No, she don't know.


Which one is correct?

Well, since it is "she", you can use either:

No, she doesn't.
No, she doesn't know.


"She do" does not exist, so you cannot use "she don't" or "she don't know".

Quick to the point, "she don't" is not the proper contraction for "she does not".

You might hear "she don't know" and not judge too harshly, but that don't [sic] make it right, when "doesn't" is so much more euphonious.
