what is, if any, the difference between these?

You have to do as I say! Period!
a) Do I?
b) I do?

There is no difference, either may be used.

I think 'Do I' is a tad more challenging than 'I do?'
'I do' seems like asking for confirmation, in a passive way.

I do? or Do I? With the question mark these are pretty much always challenging and/or sarcastic and challenging - unless said by a child - then maybe just questioning.

Learning is its own reward, and it's fun too!
