The full title of this Gosho should read, “On the Object of Worship Manifesting the True Buddha’s Entity."

Should there be a comma after 'read'?

Grammatically no. But it's common in that usage and pleasant to read.

That's right.
There are a few "styles" and conventions for what to use just before a "quotation mark" or "inverted comma" like this.

When I was at school, I was taught to use a colon - but I've also seen advices to use a comma or nothing at all.

The full title of this Gosho should read: “On the Object of Worship Manifesting the True Buddha’s Entity."

As you can see from my first sentence, if the quotation marks are just around one word, or maybe a short phrase, I prefer to use nothing.
I definitely feel that a colon is more than is necessary.
